DNA repair proteins work around nucleosomes, which package DNA in cells and limits DNA accessibility for DNA repair factors. Here, by using cryo-EM and biochemical assays, the authors show that DNA-PKcs promotes NHEJ by overcoming histone barriers and moving along nucleosomal DNA.
